CMYK to spot

I have a blue gradient with some black strokes running through it that was created in photoshop. In Quark it seperates into CMYK. I want to change the blue gradient in photoshop to PMS 300 CVU and leave the black strokes alone so that it in Quark it will seperate into 2 colors (PMS 300 and black). Does anybody know how to do this?
