Edit>Define Brush Preset

I am running CS2 on WinXP. I want to use a part of an image as a brush. No matter how I approach it, that option is greyed out under the Edit menu. How can I make it available?

Thanks anyway…I had a cranial-rectal inversion, but I am better now! The "part of the image" can be no larger than 2500 pixels. Once i made that change all works as planned.
