My system: Opteron 270 processor w/3 Gig RAM, Windows XP SP2, 400 gig free hard drive space.
I've noticed this for a while, now. After I save and close a file, leaving CS2 open, I try to email the edited file via Outlook XP - I can't email it until I shut CS2 down. Is it not unlocking the file? (Thanks.)

No, Photoshop doesn't do that.
Something else on your system is locking the files.

Hi Richard,
One way to check what is using the file is to disable all startup items and temporary disable firewall/anti spyware programs.
(to disable startup items go to start(windows)- Run - and type 'msconfig' go to the startup tab and disable all.

Thanks for all the input! I've just downloaded Process Explorer, which is supposed to have a feature that shows what processes have a file locked; so far, no luck. I've emailed the author on the failure to identify the locking process, and will update this thread if I find a solution via this method.

That could mean that the lock is due to the OS or a low level driver (anti-virus, or other utility program).
