I am using Photoshop 6 to view a grayscale TIF image.

I read that if I Edit the Color Settings for Dot Gain, the image I am viewing should show the affect of the changes.

When I Edit the Color Settings and choose a profile such as US Defaults with Dot Gain set to 30%, I do not notice any difference in the image displayed versus 10% or just setting Color Management off.

Is there something else I have to do to see the image with dot gain applied?

Color Settings affects only your default profiles. It does not change the settings for any existing image. Use Assign Profile instead, and you’ll see the change you expect.
